First you have to arrive to Buenos Aires.
From Rio de Janeiro and/or Sao Paulo to BA us about 30 to 36 hours by bus.
Then another 30 hours to Usuahia, the South-most city in the world.

I have traveled from Buenos Aires to ushuaia in a car and it took me 3 days. Depending on where in Brazil you are, I would calculate about 1 day/800 km or, if you want to drive really fast 1 day/1000 km. Good luck!
The travel in fly from Bs As to El Calafate or Ushuaia is cheaper than bus. With bus you has 36 hs sat down. It´s terrible!! There is 2900km. And the cost is the same or more that fly, although you don´t believe me. But if you go to Bariloche, something more to the north, the bus is a good option, although you will have 18 hs of travel.
The travel from San Pablo tu bs as is near 24hs, but is cheaper than fly.
